data-term-id=\"142\">immunofluorescence<\/span>?<span class=\"ez-toc-section-end\"><\/span><\/h2>\n<p>Direct immunofluorescence (DIF) is a technique used in the laboratory to diagnose diseases of the skin, kidneys, and other organ systems. It is also called a direct immune fluorescent test or <span class=\"term\" data-term-id=\"949\">primary<\/span> immunofluorescence<\/p>\n<p>DIF implies the application of <span class=\"term\" data-term-id=\"425\">antibody<\/span>&#8211;<span class=\"term\" data-term-id=\"468\">fluorophore<\/span> <span class=\"term\" data-term-id=\"470\">conjugate<\/span> <span class=\"term\" data-term-id=\"1853\">molecules<\/span> to patient tissue samples obtained from <span class=\"term\" data-term-id=\"1111\">biopsies<\/span>. These antibodies-fluorophore <span class=\"term\" data-term-id=\"471\">conjugated<\/span> abnormal target <span class=\"term\" data-term-id=\"473\">bowel movements<\/span> protein in the patient&#039;s tissue. When exposed to light, the fluorophore emits its own frequency of light, seen under a microscope. Since false negative results can arise from samples from the lower extremities, avoid these sites if possible.<\/li>\n<li>\n<strong>Connective tissue diseases<\/strong>: the biopsy should be taken from a <span class=\"term\" data-term-id=\"164\">injury<\/span> (often in areas exposed to the sun), ideally over 6 months old but still active. An additional sample is often taken in a place protected from the sun.<\/li>\n<li>\n<strong>Vasculitis<\/strong>: For best results, take a <span class=\"term\" data-term-id=\"726\">puncture biopsy<\/span> or a deep<span class=\"term\" data-term-id=\"727\">shaving biopsy<\/span> from an injury that is less than 24 hours old.<\/li>\n<\/ul>\n<p>Another routine hematoxylin-eosin (H&amp;E) biopsy is usually taken. <span class=\"term\" data-term-id=\"128\">histology<\/span>. Note that the optimal biopsy site and required time differ for these examples.<\/p>\n<ul>\n<li>\n<strong>Autoimmune bullous diseases<\/strong>: remove an integer <span class=\"term\" data-term-id=\"300\">vesicle<\/span> or biopsy of the edge of a blister.<\/li>\n<li>\n<strong>Vasculitis<\/strong>- Select a purple lesion less than 72 hours long.<\/li>\n<\/ul>\n<h2><span class=\"ez-toc-section\" id=\"Transporte-y-almacenamiento-de-la-muestra-de-biopsia\"><\/span>Transport and storage of the biopsy sample.<span class=\"ez-toc-section-end\"><\/span><\/h2>\n<p>Samples for DIF should not be placed in formalin, as this alters the proteins and significantly decreases the precision of the results. Options for transporting the sample include:<\/p>\n<ul>\n<li>Placed in a <span class=\"term\" data-term-id=\"1875\">saline<\/span>soaked gauze<\/li>\n<li>In saline solution <span class=\"term\" data-term-id=\"1119\">solution<\/span>\n<\/li>\n<li>In special media (eg, Michel or Zeus)<\/li>\n<li>Frozen in liquid nitrogen (sample should not be allowed to thaw).<\/li>\n<\/ul>\n<p>Each laboratory has its own protocols. Samples in saline solution can produce <span class=\"term\" data-term-id=\"277\">higher<\/span> sensitivity over other media if processed within 24-48 hours.<\/p>\n<h2><span class=\"ez-toc-section\" id=\"%C2%BFQue-le-sucede-a-la-muestra-en-el-laboratorio\"><\/span>What happens to the sample in the laboratory?<span class=\"ez-toc-section-end\"><\/span><\/h2>\n<p>DIF can be carried out on an automated machine or manually.
